      目的探索济川煎对慢传输型便秘( STC)大鼠的治疗作用及其对磷脂酰肌醇 3激酶( PI3K)/蛋白激酶 B(Akt)信号通路的影响。方法 2024年4—6月,将60只SD大鼠采用随机数字表法分为六组:健康对照组、模型组、阳性药组(莫沙必利 1.65 mg/kg)、济川煎高剂量组( 7.40 g/kg)、济川煎中剂量组( 3.70 g/kg)、济川煎低剂量组( 1.85 g/kg),每组 10只。除健康对照组外,其余各组灌胃盐酸洛哌丁胺建立大鼠 STC模型。造模成功后,各给药组按相应剂量灌胃,每天 1次,连续 14 d。实验结束后,观察首粒黑便排出时间、 8h粪便颗粒数、粪便含水量和炭末推进率。取肠道组织进行苏木精 -伊红( HE)染色,观察济川煎对 STC大鼠肠道组织病理变化的影响。采用酶联免疫吸附分析( ELISA)检测大鼠血清中胃动素、胃泌素和炎症因子肿瘤坏死因子 α(TNF-α)、白细胞介素( IL)-1β、IL-6的含量。采用蛋白质印迹法和实时荧光定量逆转录聚合酶链反应( qRT-PCR)检测 PI3K/Akt信号通路相关蛋白和 mRNA含量。结果健康对照组、模型组、阳性药组、济川煎高剂量组、济川煎中剂量组、济川煎低剂量组首粒黑便排出时间分别为( 377.19±17.58)min、(455.15±13.99)min、(389.44±16.74)min、(397.09±12.63)min、(418.89±17.11)min、

      Objective The aim of this study was to explore the therapeutic effect of Jichuan decoction in rats with slow transit consti. pation (STC) and its influence on the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway.Methods From April 2024 to June 2024, 60 SD rats were randomly divided into 6 groups using a random number table: healthy control group, modelgroup, positive drug group (mosapride group, 1.65 mg/kg), Jichuan decoction high-dose group (7.40 g/kg), Jichuan decoction medium-dose group (3.70 g/kg) and Jichuan decoction low-dose group (1.85 g/kg), with 10 rats in each group. Except the healthy control group,the STC model was established in the other groups by intragastric administration of loperamide hydrochloride. After successful model.ing, each treatment group was administered the appropriate dose by gavage once a day for 14 days. After the experiment, the excretiontime of the first black stool, the number of fecal pellets in 8 h, fecal water content, and the rate of carbon powder propulsion were ob.served. Hematoxylin and eosin (HE) staining was used to observe the effect of Jichuan decoction on the pathological changes of intesti. nal tissue in STC rats. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum levels of motilin (MTL), gastrin (GAS),and the inflammatory factors tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and IL-6. Western blotting and real-time quanti. tative reverse transcription polymerase chain reaction (qRT-PCR) were used to detect protein and mRNA expression levels related to the PI3K/Akt signaling pathway.Results The first black stool excretion times in the healthy control group, model group, positive drug group, Jichuan decoction high-, medium-, and low-dose groups were (377.19±17.58) min, (455.15±13.99) min, (389.44±16.74) min,(397.09±12.63)min, (418.89±17.11) min, and (446.30±15.77) min, respectively. The p-PI3K/PI3K ratios were 0.32±0.05, 1.37±0.13, 0.75±0.10, 0.66±0.08, 0.84±0.12, and 1.13±0.09, respectively, and the p-Akt/Akt ratios were 0.40±0.01, 1.15±0.05, 0.58±0.03, 0.50± 0.02, 0.72±0.03, and 1.07±0.02, respectively. Jichuan decoction improved the first black stool excretion time, fecal pellet number, fe.cal water content, and carbon powder propulsion rate in STC rats, reduced colon mucosal injury, increased the number of goblet cells,increased serum MTL and GAS levels, decreased the levels of the inflammatory factors TNF-α, IL-1β, and IL-6, and decreased the ex. pression levels of PI3K/Akt signaling pathway-related proteins and mRNA.Conclusion Jichuan decoction can improve STC symp.toms in rats, an effect that may be related to regulating gastrointestinal hormone and inflammatory factor levels and inhibiting the PI3K/Akt signaling pathway.
