      目的:探讨清心滋肾汤治疗围绝经期综合征的作用机制。方法:基于网络药理学的方法,通过检索 TCMSP 数据库获取清心滋肾汤中各药物的活性成分;通过GeneCards、PharmGkb 和 Drugbank数据库收集围绝经期综合征疾病靶点,将活性成分与疾病基因靶点取交集,得到清心滋肾汤治疗MPS的预测靶点。采用 Cytoscape 3.8.0 构建“药物-成分-靶点-疾病”网络;通过 STRING 数据库和 Cytoscape 构建蛋白互作网络(PPI)及精简核心网络;运用 R软件对清心滋肾汤治疗MPS进行 GO 富集分析和 KEGG 通路富集分析。结果:筛选出 69个主要活性成分,涉及 54个基因靶点,其中主要活性成分包括木犀草素(Luteolin)、槲皮素 (Quercetin)、豆甾醇(Stigmasterol)、谷固醇(Beta-sitosterol)、山柰酚(Kaempferol)、四氢鸭脚木碱(Tetrahydroalstonine)等;核心靶点基因包括IL6、IL1B、ESR1、SERPINE1、PPARG、CRP、CCND1、PGR、HMOX1、VCAM1、CYP19A1、IGFBP3等;富集到 GO 生物过程(BP)1180条、细胞定位(CC)39条、分子功能(MF)61条;KEGG 通路 162 条,包括糖尿病并发症中 AGE-RAGE 信号通路、脂质和动脉粥样硬化信号通路、PI3K-Akt信号通路、流体剪切应力和动脉粥样硬化通路等。结论:清心滋肾汤通过木樨草素、槲皮素、豆甾醇、谷固醇、山柰酚、四氢鸭脚木碱等有效成分作用于IL6、IL1B、ESR1、SERPINE1、PPARG、CRP、CCND1、PGR、HMOX1、VCAM1、CYP19A1、IGFBP3等关键靶点通过调节糖脂代谢、抗氧化、改善雌激素代谢等作用机制达到治疗围绝经期综合征的作用。

      Objective: To explore the mechanism of Qingxin Zishen Decoction in the treatment of perimenopausal syndrome. Methods: Based on the method of network pharmacology, the active ingredients of each drug in Qingxin Zishen Decoction were obtained by searching the TCMSP database; the perimenopausal syndrome disease targets were collected from the GeneCards, PharmGkb and Drugbank databases, and the active ingredients and the disease gene targets were collected Click the intersection to get the predictive target of Qingxin Zishen Decoction for the treatment of perimenopausal syndrome. Use Cytoscape 3.8.0 to construct a "drug-component-target-disease" network; construct a protein interaction network (PPI) and streamline the core network through STRING database and Cytoscape. Go enrichment analysis and KEGG pathway enrichment analysis of Qingxin Zishen Decoction in the treatment of MPS were carried out by R software. Results: 69 main active ingredients are screened, involving 54 gene targets, among which the main active ingredients include Luteolin, Quercetin, Stigmasterol, Beta-sitosterol, Kaempferol, Tetrahydroalstonine etc.; The core target genes include IL6,IL1B,ESR1,SERPINE1,PPARG,CRP,CCND1,PGR,HMOX1,VCAM1,CYP19A1,IGFBP3 etc.; GO enrichment items are obtained, including the 1180 Biological Process (BP) ,39 Cellular Component (CC),61 Molecular Function (MF); There are 162 KEGG pathways, including AGE-RAGE signaling pathway in diabetic complications, lipid and atherosclerosis signaling pathway, PI3K-Akt signaling pathway, fluid shear stress and atherosclerosis pathway. Conclusion: Qingxin Zishen Decoction acts on key targets like IL6, IL1B, ESR1, SERPINE1, PPARG, CRP, CCND1, PGR, HMOX1, VCAM1, CYP19A1 and IGFBP3 by regulating glycolipid metabolism, antioxidation, improving the mechanism of estrogen metabolism to treat perimenopausal syndrome.
