      目的探讨术前血清长链非编码 RNA肿瘤蛋白翻译调节因子 1-反义 RNA1(lncRNA TPT1-AS1)、整合素 β3(ITGB3)水平与结直肠癌病人根治性切除术后复发转移的相关性。方法选择 2019年 3月至 2020年 3月在海南省第二人民医院进行结直肠癌根治性切除术的 90例病人为结直肠癌组,选择同期在海南省第二人民医院进行健康体检的人员 90例作为对照组,实时荧光定量逆转录聚合酶链式反应( qRT-PCR)检测血清 lncRNA TPT1-AS1表达水平,酶联免疫吸附测定( ELISA)检测血清 ITGB3水平,搜集病人资料, Pearson法分析 lncRNA TPT1-AS1与 ITGB3水平相关性, Cox回归模型分析影响结直肠癌根治性切除术后复发转移的危险因素。结果与对照组比较,结直肠癌组病人术前血清 lncRNA TPT1-AS1表达水平( 2.62±0.35比 1.05±0.12)、 ITGB3水平[( 236.24±30.62)ng/L比( 185.47±25.38)ng/L]显著升高( P<0.05);结直肠癌病人血清 lncRNA TPT1-AS1和 ITGB3水平呈正相关性( r=0.65,P<0.05);不同分化程度、 TNM分期、浸润深度、淋巴结转移及血管侵犯的病人血清 lncRNA TPT1-AS1、ITGB3水平比较差异有统计学意义( P<0.05); lncRNA TPT1-AS1、ITGB3高表达组结直肠癌根治术后复发转移率显著高于低表达组( P<0.05); TNM分期、淋巴结转移、血管侵犯、 lncRNA TPT1-AS1、ITGB3是影响结直肠癌病人术后复发转移的危险因素( P<0.05)。结论结直肠癌病人根治性切除术前血清 lncRNA TPT1-AS1、ITGB3水平升高,与术后复发转移相关,可能成为结直肠癌预后判断的血清标志物。

      Objective To investigate the correlation between serum long non-coding RNA tumor protein translationally controlled 1antisense RNA1 (lncRNA TPT1-AS1) and integrin β3 levels and recurrence and metastasis of colorectal cancer patients after radical re‐section.Methods A total of 90 patients who underwent radical resection of colorectal cancer in the Second People's Hospital of Hainan Province from March 2019 to March 2020 were selected as the observation objects, 90 healthy persons who had physical examination in the Second People's Hospital of Hainan Province at the same time were selected as the control group. The expression level of serum lncRNA TPT1-AS1 was detected by qRT-PCR, the level of serum ITGB3 was detected by ELISA, the data of patients were collected, Pearson method was used to analyze the correlation between lncRNA TPT1-AS1 and ITGB3 levels, and Cox regression model wasused to analyze the risk factors of recurrence and metastasis of colorectal cancer after radical resection.Results Compared with the control group, the levels of serum lncRNA TPT1-AS1 expression (2.62±0.35 vs. 1.05±0.12) and ITGB3 [(236.24±30.62) ng/L vs. (185.47±25.38) ng/L] were obviously increased in colorectal cancer patients before operation (P<0.05); there was a positive correlation between serum lncRNA TPT1-AS1 and ITGB3 levels in patients with colorectal cancer (r=0.65, P<0.05); the levels of serum lncRNA TPT1-AS1 and ITGB3 in patients with different differentiation degree, TNM stage, invasion depth, lymph node metastasis and vascularinvasion were statistically obvious (P<0.05); the recurrence and metastasis rate of colorectal cancer after radical operation in the high expression lncRNA TPT1-AS1 and ITGB3 groups was obviously higher than that in the low expression groups (P<0.05); TNM stage, lymph node metastasis, vascular invasion, lncRNA TPT1-AS1, and ITGB3 were risk factors for recurrence and metastasis of colorectal cancer patients after surgery (P<0.05).Conclusion The levels of serum lncRNA TPT1-AS1 and ITGB3 in patients with colorectal cancer before radical resection are elevated, which are related to the recurrence and metastasis after operation, and may be serum markersfor the prognosis of colorectal cancer.
